Expected Usage
This Product is suitable for the collection, transportation and storage of sputum samples, The nucleic acids extracted from preserved samples are suitable for amplification and detection.
1.One 50-mL collection tube containing 3 mL preservative cocktails; sample bag.
2.Two different preservative formula of preservative cocktails:
Type A: kit with Virus Inactivation: main components include Tris, non-denatured detergent mix, EDTA, DTT and preservative cocktails.
Type B: kit with Virus Protection. HEPES, KH2PO4, KCL, NaCl, NaHCO3, Na2HPO4.12H2O, Glucose, phenol Red, and preservative cocktails
Back Ground
1. The sputum samples for the detection of respiratory tract infections is convenient and reliable, which is confirmed by recent publication. Samples collected from lower respiratory tract have a higher positive detection rate of all kinds of virus infection than other methods.
2.Sputum usually is very thick and sticky. One of the challenges of preserving sputum samples is how to effectively preserve pathogens like all kinds of virus, a single strained RNA virus, and/or pathogen nucleic acid from degradation during sample collection and transportation.
Functions and Features
Type A. Contains preservatives that inactivate pathogens such as viruses
and protects RNA/DNA samples from degradation.
The main components of preservatives include Tris buffer, nondenatured detergents, DTT and proprietary cocktails that prevent bacterial and fungal growth, as well as inactivate RNase/DNase activities. No guanidinium and other compounds that may interfere with nucleic acid extraction or PCR amplification is included. Preserved samples are stable in the room temperatures for 5 days.
Type B. Preserve pathogens intact such as all kinds of virus. The recipe is not Intended to kill the pathogens (viruses).
The main components include HBSS, HEPES, DTT, and the proprietary cocktails that stabilize the pathogen membranes, inactivateRNase/DNase activities and bacterial and fungal growth. Preserved samples can be stored up to two weeks.
Sputum Collection Method
1 .Rinse mouth and don't eat anything 30 min prior to the collection.
2.Open the Sampling kits and label the 50-mL collection tube.
(Note: the preservative solution smells and is normal)
3. Cough 2-3 times and spat the sputum into the collection tube. The total sputum will be around 1 - 2 mL.
4. Close the cap tightly and shake the tube vigorously.
5. Place the collection tube into the sample bag.
6. Pack the sample bag with another zip bag and place in the sample collection containers.
7. The collected samples canbe shipped in room temperature.
